Retatrutide (development code LY3437943) is an experimental drug from Eli Lilly. It is a single engineered peptide that activates three hormone receptors at once: GIP, GLP-1 and glucagon.
It carries a fatty-acid attachment that extends its half-life to about six days. It is investigational and has not been approved by any regulator.
Drugs acting on GLP-1 alone, or GLP-1 plus GIP, are already approved for diabetes and obesity. Retatrutide adds glucagon receptor activity, which in mice appeared to raise energy expenditure and contribute roughly a third of the weight loss.
In a 2023 phase 2 trial of 338 adults with obesity, body weight fell by an average of up to 24.2% at 48 weeks across the tested regimens, compared with 2.1% on placebo. Results from several phase 3 trials in the TRIUMPH program were announced by Lilly in 2026.

Retatrutide has unusually strong clinical evidence for an unapproved compound, including a published NEJM phase 2 trial and several large phase 3 trials. However, the phase 3 results so far come from company press releases and have not all been peer reviewed. Nearly all data come from the developer's own studies.
