Quick Summary
FDA-approved ingredient (Mounjaro 2022, Zepbound 2023)Tirzepatide is a 39-amino-acid peptide from Eli Lilly that activates two gut-hormone receptors, GIP and GLP-1, which help regulate insulin release, blood sugar and appetite. It is the active ingredient of two FDA-approved prescription medicines, Mounjaro (type 2 diabetes) and Zepbound (chronic weight management and obstructive sleep apnea). This research-grade material is neither of those approved drug products.

Why Researchers Study It
- Combining GIP and GLP-1 receptor activity in one molecule
- Blood-sugar control in type 2 diabetes (SURPASS trial program)
- Body-weight regulation in obesity (SURMOUNT trial program)
- How GIP receptor activity adds to GLP-1 effects, studied in cells, animals and people
- Long-acting peptide design through fatty-acid attachment and albumin binding
What It IsTirzepatide (development code LY3298176) is a synthetic peptide of 39 amino acids with a C20 fatty diacid attached.
Tirzepatide (development code LY3298176) is a synthetic peptide of 39 amino acids with a C20 fatty diacid attached. The fatty chain binds blood albumin, which gives it a half-life of about five days.
A single molecule activates both the GIP receptor and the GLP-1 receptor, the two main incretin (gut-hormone) receptors that help regulate insulin release after meals. It was designed by Eli Lilly starting from the GIP hormone sequence.
The molecule, the approved medicines and this product are three different things. Tirzepatide is the molecule. Mounjaro (approved May 2022 for type 2 diabetes) and Zepbound (approved November 2023 for chronic weight management, and December 2024 for moderate-to-severe obstructive sleep apnea in adults with obesity) are Lilly's FDA-approved drug products, made and tested under FDA oversight. Renuvia's tirzepatide is research-grade material for laboratory use; it is not Mounjaro or Zepbound and has not been reviewed or approved by the FDA.
The FDA has warned about unapproved tirzepatide sold online, including products labeled "research use only", and has issued warning letters to sellers of such products.
How It Works3 established · 2 proposed or under investigation
Established
- Binds and activates both the GIP and GLP-1 receptors (FDA label); in cell assays it acts more strongly at the GIP receptor than at the GLP-1 receptor (Coskun 2018)
- Increases glucose-dependent insulin release and lowers glucagon levels (FDA label)
- Half-life of about five days, due to the attached C20 fatty diacid and albumin binding (FDA label)
Proposed / under investigation
- How much GIP receptor activity contributes to weight loss beyond GLP-1 activity is still debated
- Effects on appetite and food intake are thought to act through brain GLP-1 and GIP receptors; the pathways are not fully mapped in humans
Research Status & EvidenceFDA-approved ingredient (Mounjaro 2022, Zepbound 2023)
The approvals apply only to Lilly's Mounjaro and Zepbound. This product is research-grade material, not an approved drug, and has not been evaluated by the FDA.
Types of evidence
- Available: Cell & lab studies
- Available: Animal studies
- Not available: Early human studies
- Available: Clinical trials
- Available: Approved medical use
Tirzepatide has extensive clinical evidence, including large peer-reviewed phase 3 trials and full FDA approvals. In SURMOUNT-1, adults with obesity lost substantially more body weight over 72 weeks than with placebo. That evidence applies to the approved medicines made under FDA oversight, not to research-grade material of unverified identity, purity or sterility.
Areas of research
- Type 2 diabetesapproved indication (Mounjaro), SURPASS trials
- Obesityapproved indication (Zepbound), SURMOUNT trials
- Obstructive sleep apnea with obesityapproved indication (Zepbound, 2024)
- Receptor pharmacologydual GIP / GLP-1 agonism in cell assays and animals
Limitations
- FDA approval covers only Lilly's Mounjaro and Zepbound; this research-grade product is not an approved drug and has not been evaluated by the FDA
- The FDA has warned that unapproved GLP-1 products sold online, including ones labeled "research use only", are of unknown quality and may be harmful
- The approved labels carry a boxed warning for thyroid C-cell tumors, seen in rats; whether this applies to humans is unknown
- Most common side effects in trials were gastrointestinal (nausea, diarrhea, vomiting, constipation)
